A comparison between enzyme immunoassay and HPLC for ochratoxin A detection in green, roasted and instant coffee BABT

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for ochratoxin A (OTA) detection in green, roasted and instant coffees was developed using anti-OTA monoclonal antibody. Immunological reagents prepared were OTA-BSA (4.76 mg/mL), anti-OTA.7 MAb (2x10³-fold dilution) and HRP-anti IgG (10³-fold dilution). The detection limit was 3.73 ng OTA/g and correlation coefficients (r) between this immunoassay and high performance liquid chromatography were 0.98 for green coffee, 0.98 for roasted and 0.86 for instant. OTA levels detected by ic-ELISA were higher than by HPLC, with ELISA/HPLC ratio of 0.66 - 1.46 (green coffee), 0.96 - 1.11 (roasted) and 0.93 - 1.82 (instant). Evaluation of yeast cell wall on the performance of broiles fed diets with or without mycotoxins Rev. Bras. Ciênc. Avic.

This experiment aimed at evaluating the effects of the interactions between aflatoxin (500 or 250 ppb) and ochratoxin (500 or 250 ppb), and the possible benefits of adding yeast cell wall to prevent the effects of these mycotoxins in broiler chickens. Relative organ weight gain and live performance were evaluated at 21 and 42 days of age. Results indicated that at the levels of mycotoxins included in the experimental diets, ochratoxin reduced feed intake and body weight gain, and aflatoxin only affect feed intake of 21-day-old birds. Molecular identification of Aspergillus spp. isolated from coffee beans Scientia Agricola

Some species belonging to the genus Aspergillus are potential producers of ochratoxin A (OA), a mycotoxin with nephrotoxic, immunosuppressive, teratogenic and carcinogenic effects. The aim of the present study was to identify the species of Aspergillus that contaminate the inside of coffee beans collected in the stage of maturation and drying, from 16 producing areas located in the northern region of the State of Paraná, in the South of Brazil. A total of 108 isolates of Aspergillus spp. was identified at the species level, by sequencing the internal transcribed spacer (ITS1-5.8S-ITS2) of ribosomal DNA (rDNA). Abstract Mycotoxins are secondary metabolites produced by specific filamentous fungi that are common contaminants of agricultural commodities. These compounds are toxic to humans and animals, and they are a health problem worldwide. Mycotoxin-containing feeds can cause serious diseases in farm animals and substantial economic losses. Therefore, the objective of this study was to determine the occurrence and levels of six mycotoxins in three types of forage at retail in the state of Nuevo León, México. Survey of mycotoxins in Southern Brazilian wheat and evaluation of immunoassay methods Scientia Agricola

ABSTRACT One hundred commercial wheat grain samples were collected during the 2015 sea-son across 78 municipalities in the states of Paraná (PR), Rio Grande do Sul (RS), and São Paulo (SP), Brazil. Separate subsamples were analyzed for the concentration of deoxynivalenol (DON), zearalenona (ZEA) and ochratoxin A (OTA) mycotoxins using two methods: UHPLC-MS/MS (reference method) and a commercial enzyme-linked immunosorbent assay (ELISA) (AgraQuant®). The OTA mycotoxin was not found in the samples by both methods. DON and ZEA were detected in 55 % and 39 % of the samples by the reference method, with overall mean levels of 795.2 μg kg−1 and 79.78 μg kg−1, respectively.
